I did start with "try this"
BRLTTY is used for braille keyboards so takes priority over anything else, found this out the hard way a while back where USB stuff using serial would work on some PC's and laptops, but will not work no matter what i do with permissions and groups and dialouts on some other PC's and laptops. Removed that and everything worked. Took about 2 hours of searching as the whole f@kin internet is copy/paste the same "add user to group" and "chmod a+x ttyACM0" ! Where the hell did they get ACM0? I have never ever seen that on my equipment, and i do have a lot of equipment, a lot!
I need help...

My family, friends, neighbors are all good, so yes it is, some occasional s#it at the north side, luckily last time they f@ked up badly as they were planning a full on invasion on that side, we lost 1 but the world found out they are the problem. Was not enough they started 4 f@kin wars here to figure that out.

Thanks for the suggestion.
Tried what you suggested. It would appear that brltty is not installed on my machine.
It is worth noting that the pmx485-test works fine so it would appear that the serial interface is operating properly.

I found the error that is causing the failure within the try/except. Because of the line it breaks on, I am guessing that your Port name is incorrect.
I pushed a fix for the aforementioned error. It will likely reveal that whatever you put for the port is not found. Unsure what you put, but it may be an issue with a leading or trailing / , etc.
After the buildbot does its thing you can update to grab the changes. For 2.10, the version will be v1.237.302

Genius!
The port name was indeed incorrect:
/dev/ttyusb0
instead of
/dev/ttyUSB0
It now works fine and talks to the Hypertherm OK.
