Follow this, no skipping:

After you wire and confirmed working the E-Stop and limit switches (the extreme limits that will shut the power to the drives), then check the encoders, make sure the scaling is right, and make sure they count in the right direction.
Only after having all of the above wired and tested and configured, wire a single drive:
Mesa  <=>  Drive
Aout  <=>  pin 2
Agnd  <=>  pin 4
Ena+  <=>  pin 8
Ena-  <=>  pin 9
I would wire a 1Kohm resistor to all those pins on the Mesa side first and test, the first part of schematic you posted is very strange, 22 is still unknown.
And do not change drawings/schematics.

OK, but to what is the other side connected? From what is that relay controlled? Are all 22 pins connected together?

Just a friendly warning, electronic do not have "i am sorry" buttons. You have to be sure what each and every wire does, how and where and to what it is wired, how it is controlled or what it controls, how it interacts with other electronics, etc, etc.

 All the 22's get grounded to cabinet with one wire.

OK, so that wiring should not be used anymore, just wiring the drives to Mesa should work.
See the link i posted on the procedure to wire and test.
